Epithalon (also called Epitalon or Epithalamin) is a synthetic tetrapeptide (Ala-Glu-Asp-Gly) modeled on a fragment of the bovine pineal polypeptide preparation epithalamin, developed at the St. Petersburg Institute of Bioregulation and Gerontology. In cultured human somatic cells it induces telomerase activity and elongates telomeres, and in rodent models it modulates pineal melatonin rhythms and circadian gene expression. Most published mechanistic and clinical work on epithalon comes from a single research group (Khavinson and colleagues), and independent replication outside that group is limited.
GHK is a naturally occurring human tripeptide (glycyl-L-histidyl-L-lysine) first isolated from human plasma by Pickart in 1973; it binds divalent copper with high affinity to form the GHK-Cu complex. The complex modulates the synthesis and breakdown of collagen, elastin, glycosaminoglycans, and proteoglycans, and influences metalloproteinases and their inhibitors during dermal remodeling. Genome-wide expression analyses report that GHK shifts expression of a large fraction of human genes toward a younger profile, with effects on DNA repair, antioxidant defense, and inflammation pathways. Plasma GHK concentrations decline with age, which has framed its use as a skin and connective tissue signaling peptide rather than a hormonal agent.
Nicotinamide adenine dinucleotide (NAD+) is a coenzyme present in every living cell, not a peptide. It cycles between oxidized (NAD+) and reduced (NADH) forms and serves as a hydride carrier in mitochondrial oxidative phosphorylation, as a substrate for sirtuins (SIRT1 to SIRT7), PARP enzymes involved in DNA repair, and CD38. Cellular NAD+ declines with age and in several metabolic and neurodegenerative conditions, and can be replenished either directly (intravenous NAD+) or via precursors such as nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN).
