TB-500 is a synthetic peptide derived from thymosin beta-4 (Tβ4), a 43 amino acid actin-sequestering protein that is one of the most abundant intracellular proteins in mammalian cells. Tβ4 binds G-actin and regulates the actin cytoskeleton, and through this and related activities it promotes cell migration, angiogenesis, re-epithelialization, and suppression of inflammation in models of tissue injury. The commercial “TB-500” sold as a research peptide is commonly described as the active N-terminal actin-binding fragment of Tβ4 rather than the full-length 43mer; published clinical trials have used full-length synthetic Tβ4 (RGN-259, RGN-352) rather than the truncated TB-500 sequence.
KPV (lysine-proline-valine) is a tripeptide corresponding to the C-terminal residues 11 to 13 of alpha-melanocyte stimulating hormone (alpha-MSH). Published cell and animal work indicates KPV exerts anti-inflammatory activity by entering intestinal epithelial and immune cells through the di- and tripeptide transporter PepT1, then inhibiting NF-kB and MAP kinase signaling and reducing transcription of pro-inflammatory cytokines such as TNF-alpha, IL-6 and IL-8. Unlike longer alpha-MSH analogs, the KPV fragment retains anti-inflammatory effects with limited classical melanocortin receptor agonism, suggesting both receptor independent (PepT1 mediated) and receptor dependent contributions.
BPC-157 (Body Protection Compound 157) is a synthetic pentadecapeptide (sequence GEPPPGKPADDAGLV) derived from a protective fragment of human gastric juice protein, stable in acidic gastric environments. Across rodent models it accelerates tissue repair through upregulation of vascular endothelial growth factor (VEGF), modulation of the nitric oxide (NO) system, and promotion of angiogenesis and fibroblast migration. Reported activity is largely confined to preclinical models; human clinical data remain limited to early Phase I/II work for inflammatory bowel disease conducted under the development code PL 14736.
